Pharmacokinetic modeling III
In the pharmaceutical praxis and the drug manufacturing often appears the need to modify the content of auxiliary substances. It is known that such a modification can significantly influence the drug concentration in the blood, bioavailability and the therapeutic effect. In relation to that the natural question may appear: How significant that influence may be? The paper describes a possible system-based approach to finding an answer to this question based the so-called sensitivity analysis. At the same time the paper analyses conditions under which the compartmental model and the related in-silico experiment may be considered as a surrogate of the in-vivo experiment.
Keywords: compartment model, absorption, parametric sensitivity, surrogate of in-vivo experiment
